This antibody recognizes a protein of 35-38 kDa, which is identified as Arginase 1 (ARG1). Arginase is a manganese metallo-enzyme that catalyzes the hydrolysis of arginine to generate ornithine and urea. Arginase I and II are isoenzymes which differ in subcellular localization, regulation, and possibly function. Arginase I is a cytosolic enzyme, which is expressed mainly in the liver as part of the urea cycle, whereas arginase II is a mitochondrial protein found in a variety of tissues. Keratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I keratin (keratins 9 to 23) and one type II keratin (keratins 1 to 8). Myoglobin is a cytosolic oxygen-binding protein responsible for the storage and diffusion of oxygen within myocytes. Expression of myoglobin is highest in skeletal and cardiac muscle. Myoglobin is well accepted as an O2-storage protein in muscle, capable of releasing O2 during periods of hypoxia or anoxia. In combination with other striated muscle markers such as vimentin and myogenin, myoglobin is helpful in the identification of rhabdomyosarcoma and tumors with skeletal muscle differentiation. Complement C1q A Chain (C1qa) is a subunit of C1q. C1q is composed of 6 polypeptide chains each of C1qa, C1qb, and C1qc. C1q is a component of the classical complement pathway, and thereby plays an important role in the immune response. Each C1q subunit contains an N-terminal collagen-like region and a C-terminal C1q globular domain. The presence of receptors for C1q on effector cells modulates its activity, which may be antibody-dependent or independent. Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. Alpha actinin is an actin-binding protein with multiple roles in different cell types. In non-muscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. Mammaglobin is a 93 amino acid glycoprotein with homology to other secretoglobin-uteroglobin family members. The metal-responsive element (MRE)-binding transcription factor (MTF-1) stimulates the expression of metallothioneins in response to the exposure of cells to heavy metals. MTF-1 contains six zinc fingers in the DNA binding domain. The phosphorylation of MTF-1 in response to metal exposure appears to play a significant role in the ability of MTF-1 to activate metallothionein transcription. In addition to its role in metallothionein activation, MTF-1 is involved in a post-transcription regulatory complex for ribosomal protein S25. The BRCA 1 Associated Protein 1 (BAP1) protein belongs to the ubiquitin C-terminal hydrolase subfamily of de-ubiquitination enzymes that are involved in the removal of ubiquitin from proteins. This antibody recognizes a protein of 62 kDa, identified as PAX8. It is a member of the paired box (PAX) family of transcription factors. This nuclear protein is involved in thyroid follicular cell development and expression of thyroid-specific genes. Mutations in this gene have been associated with thyroid dysgenesis, thyroid follicular carcinomas, and atypical thyroid adenomas. PAX-8 is expressed in the thyroid (and associated carcinomas), non-ciliated mucosal cells of the fallopian tubes, and simple ovarian inclusion cysts, but not normal ovarian surface epithelial cells.
